ABSTRACT
The influence of extract Strychnos ligustrina Blume wood aqueous given by
multiple doses against Plasmodium berghei in vivo
S. ligustrina Bl. is one of Loganiaceae family plants, is empirically
used as antimalarial drug in Lombok Island. Recently study has been proving that
aqueous extract of S. ligustrina Bl. possess an antimalarial activity against P.
berghei in vivo in mice. The ED50 value is 0,45mg/kg body weight given by single
dose. A single dose extract only inhibited parasite, and couldn’t increase the survival
of mice after extract has stop given. The aim of this research is to find out the effect
of aqueous extract of S. ligustrina Bl. given twice per day (multiple dose) in inhibit
parasite growth and increasing the survival of P. berghei-mice.
The study was carried out by using in vivo model which is a modification of
the “4 Day Suppressive Test” originally described by Peter. Briefly mice were
inoculated intraperitoneally with parasite (P. berghei). Extract was dissolved in
CMC-Na 0,5%, then
administered daily from initial day after infection for 7 days twice per day by
oral route. Antimalarial effect was determined by microscopic examination of thin
blood smear stain with giemsa. The blood smear was made daily from mice tail until
9 days, parasitized red blood cells was counted per 5000 erythrocytes.
Research showed that extract given twice per day able to inhibit parasite
growth (P. berghei) up to 7 days after treatment is stopped and further examine up to
day 9, showed that there is no increase in parasitemia so extract could increased the
survival of mice. The ED50 value is 0,41mg/kg body weight.
