Introduction
What is Melanoma?
Melanoma is the most severe type of skin cancer that affects
an estimated 100,000 people worldwide per year. It causes malignant tumors in the
body. Melanoma is for the most part fatal with 94% of people having a 8.5 month
survival rate,while the remaining 6% have a 5 year average survival rate.
Nude Mice
Nude mice are genetic mutants that lack a thymus gland,
because of this they have a weak immune system and a very low number of T
cells.The mice have no hair on any part of their body whatsoever which is why
they are called “nude” mice. Nude mice are used in research because their bodies
do not reject any types of tissue and tumor grafts given to them.

How Both Are Related
In this experiment, nude mice are injected with human
Melanoma cells to provoke tumors in them.The mice are later treated with
antibodies labeled with 188-Re (Rhenium) in hopes of increasing the survival rate
of the illness.
Things Done So Far
To start the experiment:

The mice were weighed and blood samples were taken from them.

Blood cells (leukocytes and platelets) in the samples were counted and the numbers were
recorded.

Human Melanoma cells were injected into the mice in order to induce tumors in them.

The mice were later treated with radio labeled antibodies to see how it would affect the tumors.

Eight days later were weighed again and more blood samples were taken from them.

Blood cell numbers were counted and recorded again.
In The Past 3 Days:

The mice were weighed and more blood samples were taken from them. The blood samples
which would be used to count leukocytes (White Blood Cells) were mixed with 2% acetic acid
so that only leukocytes could be seen,while those that would be used to count platelets were
mixed with Ammonium Oxide so that only platelets could be seen.

The number of leukocytes (WBC) and platelets in the samples were counted using a
hemocytometer,which is a slide for the microscope that has small boxes for counting cells,and
compared to the numbers recorded from the previous two times.
Desired Results
In General The Desired Long-term Results Are:
To Decrease the size of the tumors in the mice.
To Slow down the growth of the tumors,or stop their growth
altogether.
Materials Used

Cages->Used To Hold The Mice

Pipettes->Pick up liquid in Mkl(Micro liter) measurement

Pipette Tips-> The piece places at the tip of the pipette that actually holds the
liquid.

Skin Caliper-> Used to measure the size of the tumors.

Latex Gloves-> Hand Protection

Scalpels-> To cut the mice so that blood may be drawn from them.

Microscope-> Used to view cells

Hemocytometer->Slide used to view and count cells under a microscope.

2% Acetic Acid->Used to kill all other organisms in a blood sample besides
Leukocytes.

Ammonium Oxide-> Used to kill all other cells in a blood sample besides Platelets.
