ABSTRACT
The Effect of Activator and Inhibitor on the Activity of Proteolytic Enzymes
from Thermostable Bacteria Isolate of Lapindo Mud
This study reveals the difference of the effect of activator and inhibitor on the
activity of proteolytic enzyme from thermostable bacteria Isolate of Lapindo Mud. In
the preliminary research sample were taken from four different points, and eight
isolates were obtained. Then, the proteolytic enzymes was screened using a medium
of skim milk. From the eight isolates, three isolates were found to have highest
proteolytic index. Optimizing the growth of bacterias and production of proteolytic
enzymes have been done before crude enzymes was gained to test their activity
using a Lowry Method. The result of testing activity indicated that there was a
significance difference among the three isolates thermostable bacteria of the Lapindo
mud samples by the treatment of inorganic ion activator (Zn2+, Ca2+, and Mn2+) and
inhibitor (EDTA, Hg2+ dan Pb2+) concentration 0,01 M.
