Abstract:Living ring-opening polymerization (ROP) is utilized for controlled
synthesis of polymeric materials and for incorporating therapeutics agents to
polymers for various biomedical applications. Two ROP related topics will be
covered in this presentation. First, ROP of amino acid N-carboxyanhydrides
(NCAs) for the controlled synthesis of linear and branched polypeptides will
be discussed. Controlled ROP of NCAs can be readily achieved via the use of Ntrimethylsilyl amines. This new method allows for the synthesis of polypeptides
with unprecedented simplicity with precisely controlled molecular weights and
narrow molecular weight distributions. By controlling polypeptide side-chains,
we developed ionic helical polypeptides that are capable of delivering DNA and
siRNA effectively into cells. In a separate study, we developed polymerdrug conjugated nanoparticles via drug-initiated, controlled ring-opening
polymerizations followed by nanoprecipitation. This process allows for
successful formulations of sub-100 nm polymeric nanoconjugates with welldefined drug loading, quantitative drug loading efficiency and controlled release
profiles. These new type of nanoparticles are promising delivery vehicles for
targeted disease treatment with improved efficacy and reduced toxicity.
