Please, give input on:
1. Medication effects (present both in children and adults)
- related to comorbidity?
(CT effect frontal/temporal mainly in adult OCD with comorbid anxiety)
- related to disease duration?
(but AO plays not role in CT in adult OCD)
- status related to disease severity?
(but severity mainly related to SA, not CT)
2. yes/no correction for total CT/SA or average CT/SA?
Please, give input on:
3. Option to:
- combine SA and CT in order to compare with VBM
results of OCD Brain Imaging Consortium (OBIC) (de
Wit et al. 2014)?
- run tensor-based FreeSurfer, in order to compare with
FreeSurfer results OBIC (de Fouche et al. 2016)? > we
than need the raw data of all sites
- run VBM (on raw data, as we did in OBIC)

Available data:
TBSS-based FA of 19 bilateral and 5 unilateral white matter
labels (plus average FA) based on the John Hopkins University –

Effect size estimates (Cohen’s d) computed using age, sex, age
by sex interaction, age2 and age2 by sex interaction as
covariates since linear and non-linear age and sex interactions
have been reported for FA (Kochunov et al., 2012)

ENIGMA-OCD Structural Covariance (Kwon JS and Yun JY et al.): 2016/AUG/25
1. Participants: meta-analytical approach
•
•
Comprised of a total of 37 dataset (31/37 included for now)
Additional information needs to be gathered from other 6 dataset
o cortical-subcortical id match needed (Reddy, Fitzgerald)
o subcortical data available? (Nurmi)
Structural Covariance
CONFIDENTIAL
ENIGMA-OCD Structural Covariance (Kwon JS and Yun JY et al.): 2016/AUG/25
2. Construction of structural covariance matrix
• ROI: 33 CSAM + 33 CTM + 6 subcort_volM  72 ROIs
 Exclusion of entorhinal cortices & nucleus accumbens (poor parcellation)
• Inclusion criteria for participants for each study sites (for meta-)
‘or(isnan(ROIL)==0, isnan(ROIR)==0)==1’ for all of the 72 ROIs, so that
computation of ROIM(mean) value available
• To regress-out effect of age, sex and ICV/mean CT/whole CSA
• Z-score transform the ‘r’ using MHC/SDHC of given ROIM
• Calculation of individualized structural covariance matrix
 1./exp((X(i,j)-X(i,k))^2)
Yun JY and Kwon JS et al., (2015, 2016)
3. Graph theory analysis
• For adults & pediatric dataset
• Global characteristics (K=0.10-0.25): compare AUC values
• Regional characteristics; community structure; rich-club phenomena
ENIGMA-OCD Structural Covariance (Kwon JS and Yun JY et al.): 2016/AUG/25
4. Meta-analytic integration
CONFIDENTIAL
4-1. Global characteristics in adult (OCD vs. HC)
(other results to be shared in next TC)
