Workflow
 Sample preparation (e.g. cells treated with different
compounds)
 Cell lysis
 Mixing samples with target gene specific capture and
reporter RNA probes
 Hybridization at 65°C over night
 Removal of non-hybridized RNA probes by nCounter
SPRINT fluidic system
 Single molecule quantification of captured RNA via
gene specific barcodes
 Advantages
 Up to 800 genes profiled in parallel
 Up to 36 samples/day
 Ultra high precision (as low as 10% change in
expression levels)
 Applicable with purified RNA and non-purified RNA
(cell lysates)
 Only 50ng RNA required (ideally 5ul 10 ng/µl)
 Pre-selected gene panels available (e.g. pan cancer,
pan cancer immune, pan cancer progression,
inflammation etc.)
