ABSTRACT
STANDARD PARAMETER DETERMINATION OF RAW MATERIAL AND
ETHANOL EXTRACT 96 % TURMERIC
RHIZOME (Curcuma domestica Val.)
Dyah Handayani
Turmeric (Curcuma domestica Val.) as medicinal plant found in almost
Indonesia region, have been used as traditional medicine. Turmeric contains 3-5 %
yellow pigment which is called curcuminoid. Curcuminoid in turmeric consist of
curcumin, monodesmetoksicurcumin, bidesmetoksicurcumin. So far, in its uses,
Turmeric will be developed as standarized herbal drug and phytopharmaca
product. Phytopharmaca as pharmaceuticals product have to comply qualification of
modern drug, that is standarized finished product and raw material. Standard
parameter of raw material and extract which will be used, have to conducted before to
get standarized product. The aim of this study is to determine spesific parameter and
nonspesific parameter of raw material and extract turmeric rhizome.
Turmeric rhizome which have been harvested, was washed, cuted, dried in
open air and grinded into powders. In this stage, spesific parameters and nonspesific
parameters determination of raw material are conducted. Rhizome powder made into
extract by meceration in ethanol 96 %. The obtained extract are condensed by
rotavapor and dried with 5 % mixture of Cab-O-Sil and Avicel 1:1. Standard
parameter of extract are conducted.
The result showed non-spesific parameters of raw material were total ash was
(11,31 ± 0,08)%, acid insoluble ash was (6,89 ± 0,20)%, water soluble ash was (1,47
± 0,16)%, water content was (10,21± 0,20)%, volatile oil content was (3,93 ± 0,23)%
and loss on drying was (16,16 ± 0,28)%. The spesific parameters of raw material
showed that water soluble extractive substance was (20,71± 0,51)%, alcohol
soluble extractive substance was 13,49 ± 0.11)%, and curcuminoid content was
(5,79 ± 0,51). The spesific parameters of raw material also examined macroscopic
and microscopic of raw material.
The result non-spesific parameters ethanol extract showed that loss on drying
was (13,67 ± 0,50)%, Cu content was (1,479 ± 0.034) ppm, Zn content was (1,0335 ±
0,0410) ppm, )%, Pb content was (0,4499 ± 0,2158) ppm, positif in DDT
(qualitatively), and negatif from microbial impurities. The spesific parameters of
ethanol extract show that curcuminoid content was (23,77 ± 1,15)%. The spesific
parameters of ethanol extract also determined by general appearance of dried extract.
