ABSTRACT
Determination Pharmacokinetics of a Single 500-Miligram Oral Dose of
Levofloxacin By Taking Blood Samples in Healthy Subjects
The research to determine the pharmacokinetics profile of once daily oral
levofloxacin in seven healthy male volunteers were investigated. The aim of the study
was to know how pharmacokinetic parameters and pharmacokinetics profile in
healthy Indonesian volunteers receiving a single oral 500-mg dose of
levofloxacin. Sample were taken minimally 3 times the drug‘s half life (which was
24hours). Blood samples were serially collected at 0, 0.5, 0.75, 1, 1.25, 1.5, 2, 3, 4, 5,
8,and
24 hours. Plasma levofloxacin concentrations were assayed by
spectrofluorometry at excitation wavelength 295 nm and emission wavelength 496
nm. Pharmacokinetics analysis was performed assuming a two compartment open
model.
The pharmacokinetics profiles of levofloxacin 500 mg in blood data such as
AUC0-24 were obtained in the range of 52,74- 88,31 mg. hour/L, AUC0-∞ were
obtained in the range of 54,41-69,44 mg.hour/L, Cmaks were obtained in the
range of 8,60-10,80 mg/L, tmaks were obtained in the range of 0,50-1,25 hour, β
were obtained in the range of 0,08-0,14 hour-1, t1/2β were obtained in the range of
5,029,06 hour, α were obtained in the range of 0,38-3,16 hour-1, Cl were obtained in
the range of 5,66-9,19 L/hour, Vd were obtained in the range of 64,73-93,84 L.
