Dose Dependant Swim Assay
• Goal:
– To measure the chemotactic response of bacteria
to differing concentrations of toluene.
• Procedure:
– Plate a small number of bacteria onto center of a
swim plates containing differing concentrations of
toluene.
– Measure the distance that the bacteria
swim/grow in a 14 hour incubation period.
Problems
• Agar at the .15% concentration is EXTREMELY
fluid. We could not invert plates for incubation
or measurement/photgraphing.
• Toluene is flammable and volatile.
• Plating a consistent number of bacteria onto
the swim plate is difficult with an inoculating
loop

Further Research
• Use .3% agar (I calculated incorrectly)
• Grow the colonies out in media, take an
absorbance reading, and plate a small amount
of a dilution to achieve a similar initial cell
count among plates.
• Larger # in order to verify results which do not
agree with paper we used as guide.
• Higher/Lower Concentrations
• Gradiants
