Culturing Bacteria
Lab Preparation
 In this lab you will be designing an experiment to
test the effectiveness of a product that contains
an antibacterial chemical.
Question
 What are you going to test in this experiment?
 Make an INFORMED prediction.
 To be INFORMED you need background info:
- Ingredients in each product
-Claims made by the manufacturer
Must be written in IF…THEN… format
IF (Manipulated variable)
THEN (Responding variable)
Manipulated, Responding, Controlled variables
Experimental control group vs test
group
Preparing Petri dish
 Divide the BOTTOM of the dish into 3 or 4
sections depending on the number of products
you are testing.
 Write your initials and class period on the SIDE of
the LID.
Swabbing a sample
 Use the sterile swab and slide it across the
sample area 2-3 times.
If you are testing:
 Hand sanitizer: swab your fingers/hands
 Mouthwash: swab the inside of your cheeks
 Disinfectant: swab a surface that you would use
the disinfectant on
Create the LAWN
 Gently rub the swab across the surface of your
agar
Storing your Petri dish
 Tape your lid down
 Place in incubator
Zone of inhibition
 The size of the ZONE determines the
effectiveness of the antibiotic.
