False-negative stool occult blood tests caused by
ingestion of ascorbic acid (vitamin C). Ann Intern Med.
1975 Dec;83(6):824-826.
D.K. w/ unexplained anemia, ascorbic acid ingestion & 472 negative stool
occult blood tests. Off ascorbic acid, her stools became strongly reactive
("4+") by 3 tests for occult blood; association observed repeatedly. A new test
developed to measure stool occult blood in the presence of ascorbic acid
remained reactive throughout observation… Current tests for occult blood
depend on the pseudoperoxidase activity of heme & are inhibited by low
levels of ascorbic acid. Reducing substances chemically similar to ascorbic
acid also inhibits occult blood tests; oxidized ascorbic acid and sulfhydryl
reducing agents do not inhibit them at physiologic levels.

blood loss… maybe test is wrong. More precise
tests are more sensitive (fewer false positives), specific (fewer
false negatives) & predictive(more true positives).

“Administration of D-penicillamine & lathyrogens
such as ß-amino-propionitrile markedly alters connective tissue by preventing normal cross-linkage of
elastin & collagen…. ß-aminopropionitrile blocks cross-linkage of elastin & collagen by preventing
initial step in cross-linkage: Lysine in peptide linkage converts to amino adipic-semialdehyde.
We show that penicillamine acts after initial step, causing accumulation of elastin rich in amino adipicsemialdehyde”.
My thesis worked out d-penicillamine’s mechanism of action in blocking elastin & collagen
